I've had a similar, if not the same, problem. I mount my iso (the game Prototype) then the game's starting box loads (with the options to play, uninstall, support, quit, etc.) When I click play, it starts loading, then says
"wrong disc inserted.
Please insert the original 'prototype' CD/DVD."
with the options to "launch analysis", "retry", or "cancel". If I click retry, it shows the same message.
I am running windows 7, if that's important. I got two DVDs from a friend with one them Prototype in 11 pieces, 6 on the first disc, 5 on the other. I combined the pieces into an iso file with 7-zip, then extracted the installer so I could install it (also with 7-zip). I had some trouble getting Daemontools to work (the whole windows 7/sptd/daemontools gig), but it seems to be working fine now.
If any more info is needed to help me, just ask.
